Setting screen brightness

I've had lots of problems calibrating my screen to the point that I have an Ips panel turning up tomorrow which has max brightness output of 400 cd/m2 output. Should I set it at 30% brightness and then calibrate it? or am I completely barking up the wrong tree?
 
I've just calibrated my laptop, and found that 50% was good for me. the monitor profiling was done pretty quickly with a colormunki, so it's easy to redo to get your min / max brightness at a good level for you.
if there is somebody knowledgable on screen / printer profiling, i also have a few questions...
 
Yes it's never explained to well is it! Even the grey scale targets can be a little confusing if you can't get your screen close. Doesn't matter how many times i read how to calibrate my screen I always feel i'm missing something.
