If I take a photo and it's saved as a RAW file and I later notice when post processing in Lightroom 4 that there is an obvious spec of dust on the lens which has been picked up, how would I remove this?

I know how to remove it as a .jpg in Photoshop. Would I have to export the RAW to jpeg first and then edit the jpeg?

No don't have to export just go to develop and I think it's the 2nd icon accross it looks like a little circle I'm not at a pc so can't remember the exact name it's most probably called spot removal lol
 
It won't be dust on the lens, they are so far out of focus they're invisible. It's probably dust on the sensor. That usually shows up on areas of plain tone, like a sky, shot at a high f/number that pulls them into sharp focus. At lower f/numbers, they will disappear too.
 
agreed- maybe a sensor clean should be your next step, as these " Dust Bunnies" as they are sometimes refered to, will not go away by themselves :thumbs:
