Question on Live View focusing

When does looking through Live View seem like the subject is sharp in focus, but when you take a photo it isn't?
 
Manual or Live View AF? In Live View the point you focused on should be sharp, to focus enlarge the area you want to be sharp and then adjust.

How are you comparing?

There shouldn't be any error apart from user when using Live vIew as the image you are viewing is exactly what will be captured by the sensor.
 
Are you using a slow shutter speed?
Are you using a tripod?
If yes, it could be that just by pushing the shutter button you are creating blur. The camera can still move even on the sturdiest tripod.
 
Approx 1/80th of a second. ISO is rather high, too high in my opinion.

That's a weird bunch of settings:
Why 6.3? For it to be sharp front to back it needed smaller, you'd have got better with different framing and a larger aperture.
Why 1/80th? Too slow for sharp handheld
ISO 2500? Your camera will go much higher if you need it, don't be scared to use the higher ISO's.

I'm not even going there on what the point of the shot was,
