First duplicate the background layer. If you have the layer pallet open right click on the background and select Duplicate. Now turn off the background . Using whatever tools you like select the background area you want to turn transparent. Now go to Edit>Clear. This will now give you a clear background. . Now to be on the safe side I tend to the go to Select> Save Selection. Now you save the image I'd tend to use a PSD file as all the details are saved.
Duplicate the layer.
Turn off the background layer.
On the duplicated layer make a selection of what you wish to clear (Magic Wand is normally pretty good), then Edit > Clear.
File > Save As... and choose PNG rather than PSD or Jpeg as the format, name it, and that should be it.
