I use in-camera sharpening most of the time, and work from the JPEGs. I often shoot JPEG and Raw together as a precaution, but normally only work from the Raw when I'm not certain of the shooting conditions, especially white balance under artificial light, or when I am after a particular effect that the camera is not programmed to deliver.
Otherwise, why spend time post-processing Raws when the camera does it perfectly well for you? However, I have spent a fair bit of time trying the various Picture Styles so I know reasonably well what the camera will produce. My custom settings are very close to the defaults though!
