Check you hace AF switched on, on the lens and if its anything like the Nikon at work there is also a switch on the camera itself the also need turning on.
This happened to be me the other day and I was stressing for ages. Ended up being the lens was not seated properly on the camera. Somehow it had twisted very slightly and so the contacts weren't lined up. I was absolutely bladdered at the time ha ha so I'd probably pressed the lens release button for some stupid reason.
Take ur lens off, make sure all the contacts are there and look ok, and put it back on.
What are you trying to focus on and in what conditions?
If it's too dark, if there's little contrast in the subject or if you're too close to the subject for your lens it can all prevent you from obtaining focus.
A little more information about the circumstances will help people be more specific with their replies
