When should I use these lens's

The main lens I use is a Canon EF-S 55-250mm F4-5.6 IS II... I also have a Canon 50mm 1.8 and a 18-55mm kit lens... When should I use each one and where?

50mm for portraits, other two as a walk about. Also 55-250 when you want you need the zoom to capture something far away.
 
I find my prime is hardly ever off my camera, you will probably notice that the 50mm is your sharpest lens in terms of quality, it will be your fastest lens in terms of light.
It will be a great lens for taking photos of flowers and other close up work, if your a photoshop user you can stich some images together to achieve wider angles, it may be worth googling brenizer method for stitching a few images together with large depth of fields.

You do have a nice spread of focal lengths, you just need to learn when each lens is appropriate.
