Struggling to get the settings right with my combo of 120-300 f2.8 and Nikon D300
Seem to be missing the focus point when shooting, any pointers would be welcome.

PP is done on picassa which always looks completely different when i post here,

I think Mark (Demilion) has or had that lens as well.
 
Those look like heavy crops. Add that to the fact that your ISO is too high and that's where the problems start.

I also think that they're over sharpened.

Try using f/4 unless you absolutely have to drop to f/2.8 (which you don't here).

With a crop camera and its larger assumed depth of field i would 100% disagree and shoot at f/2.8 to give more subject isolation, Nos 1+4 in the second set clearly show too much DOF and not enough isolation and those were already shot at f/2.8

You need to understand that when possible shoot the lowest ISO possible, the 4th in the second set for example is shot at ISO 640 with a SS of 1/8000th which is maxed out on the D300, youre shooting in AV mode so if it had got brighter you would have had nowhere to go beyond the 1/8000th so you would have ended up with exposure issues, it looks slightly over exposed already.
