I've been scanning to DNG in Vuescan since it works nicely with Lightroom presets. However, while you can set the TIFF compression to "on" in Vuescan (do not use "auto" - that's another rant), it does not compress the image in a DNG. Luckily, after importing the images to Lightroom, you can take advantage of a nice feature to recover about 40 percent of the disk space. Simply highlight your scans in the library and then use the Metadata menu item "Update DNG preview and metadata" and Lightroom will work its magic and compress the DNGs. My 6x4.5 scans are going from a typical 200MB down to 130MB!
