Very jealous, New York is one of my favourite cities.
If its your 1st visit try and;
Go up the Empire State Building and the Rockefeller Centre (I'd say one in the day and one in the evening)
Get lost wondering round Central Park
Visit ground zero
Walk across the Brooklyn Bridge
Take the Staten Island ferry
Visit the boat ride out to the Statue of Liberty
Have a wander round Times Square and pick up some Broadway tickets whilst you're at it
I could go on.....New York is a photographers dream.

We're here right now.. gotta love complimentary wi-fi!!
So far we've done the Top of the Rock, Time Square (day & night!), ground zero, Battery Park, Wall Street & Liberty Island! The memory card is pretty full. Tomorrow I'm sneaking off to the Flatiron building whilst my wife destroys the credit card in Macy's!!
My only negative.. it's a Jewish holiday so B&H Photography is shut!! :'o(

Oops, forgot to mention Wall St/Battery Park.
If you want a museum, the Guggenheim is worth a visit, can be done at the same time as Central Park.

I was just about to suggest that. Start off at the top end seeing as you're near there anyway ("The High Line is located on Manhattan's West Side. It runs from Gansevoort Street in the Meatpacking District to West 34th Street, between 10th & 11th Avenues.") and aim to do it so that you will be near the bottom as the sun is going down to get the reflected light off the buildings and then some elevated street shots as the car lights come on! You won't be disappointed.
You can get an extra hour or two out of it if you suggest to your wife to meet you in China Town for dinner!
Also, Central Park around the boating lake for the autumn colour reflections. And the upper west side of the park for colours especially as the light fades.
