It strikes me that 100-400 is best for you as it gives you the option of animals one minute and birds the next. My own choice would perhaps be Canon 400 F4 DOISUSM which can comfortably handle all situations. I have one for sale but I guess its beyond you budget - if you can hire OK.
Good points made already! I could never contemplate walking many miles with a heavy tripod and 500 f4 on my back - a friend regularly does but he is 10 years younger, has no back or knee trouble, is about 5 stone heavier and a good 6 inches taller. Although he has them he rarely uses extenders...

I have been using the Sigma 150-500 for some time now along with Canon 400 F4.
Sigma will not quite match the Canon lenses but on holiday when a 150-500 range becomes an advantage AND the lighter weight I take the Sigma every time.
